I assume OP is referring to iOS games so in that case I'd have to say Infinity Blade II. Probably around 50hrs. and I still haven't finished. But in general without a doubt it would have to be the dreaded Monster Hunter Freedom Unite! 450+ hours! Ahh, good times...
LOL... i've had 1500+ hours on Unite alone. Been ultra active on GFaqs, created guides and videos on youtube, and contributed on the now dead website Reign-of-the-Rathalos. Then i played MHP3 for about 1000+ hours, until my PSP got ignored a bit, and finally forgotten. Now all my adhoc friends bought 3DS just to buy the latest MH title.

NOVA 3 - Possibly the best IOS multiplayer I've ever seen! Hoggy - Actually an extremely fun, puzzling, unique free app. Took up a lot of my time, and it isn't a joke about our TA user Hoggy
Monsters Ate My Condo - app ate up my freetime, isn't on my phone anymore tho Super Hexagon - tons of playing time and I still suck at it Happy Street, Tiny Towers, Pocket Planes - loads of hours into these, currently only play Happy Street tho. Punch Quest - must....keep.....punching Zen Pinball - best pinball app IMO, have alot of the Marvel tables but the others are great too! Tiny Wings, Jetpack Joyride, Hook World, Monster Dash, Ski Safari, Temple Run, Blot, Robot Unicorn Attack - I've put alot of time in these and a bunch of other runners Llamasoft games - Minotron, Minotaur Rescue and Goatup mainly Battleheart - this one still beats all of those that try to imitate HomeRun Battle 2 - don't play anymore but used to play TONS Mikey Shorts - MUST GET PERFECT RUNS
That's pretty impressive. But I must say that I was able to reach the second to last Gstar all on my own, just want to out that out there on the interwebz. P.S. 1000+ hrs = EPIC
Stardom: Hollywood, Simpsons: Tapped out, Drag Racing, Dream House days and Game Dev. These games are the cause of my sleepless nights. Been playing this games in the wee hours of the morning.
Probably Jetpack Joyride - there's so much stuff to unlock in that game, and earning coins to buy everything takes a lot of work!
I hadn't completed an iOS game until Ridiculous Fishing. I am entirely in love with it. I've sunk ... so much time into this since it came out. The concept alone makes me warm inside.
